Iron: the new cancer killer?

Monday April 6, 2009 (foodconsumer.org) -- According to a study conducted by scientists of the German Research Center (DFKZ), a new discovery has been made regarding the effects of iron on cancer cells. High levels of iron released in lymphoma cells stimulate free oxygen radicals, which ultimately can lead to the death of the cancer cell entirely.

Of course, it’s not as cut and dried as it seems. Chinese Medicine News is reporting that German researchers have seemingly discovered a way to “trick” cancer cells into snuffing themselves out.

Because of their voracious ability to grow and divide, cancer cells need more energy than do healthy cells. Out of this need, cancerous cells metabolically run at full throttle.

And herein lies their ultimate demise, when it comes to this potential treatment. To satiate cancer cells, large amounts of micro-nutrients, such as iron, are required. By selectively introducing a high volume of iron into the cells, scientists have stimulated the release of harmful free radicals. The ever ravenous cancer cell in turn tries to protect itself against said radicals by inactivating them through a process of binding them to iron storage proteins, causing oxidative stress.

This oxidative stress damages molecules and genes within the cancer cells, ultimately inducing their death. DFKZ researchers teamed up with physicians of the Dermatology Department of Mannheim University Hospitals to study this process in treating a particularly aggressive form of lymphoma, called Sezary’s disease. Additionally, they have found that this treatment is also helpful in fighting other types of lymphomas that are impervious to other forms of treatment.

Couldn’t this same process destroy healthy cells just as easily as it kills the cancerous ones? Certainly, but by “selectively” injecting the cells with the iron, the healthy cells are left unscathed throughout the entire procedure.

However, the relationship between dietary iron and cancer risk in individuals without hemochromatosis is less clear. Several epidemiological studies have reported associations between measures of increased iron status and the incidence of colorectal cancer or the occurrence of precancerous polyps (adenomas), but the associations were not consistent. Biophysical concepts. Crystallization of iron (generation of ‘ferromagnetosomes’) is ordinary process into any cell (human, animal, vegetative). Some people called it a virus, some a fungus, one called it a mould, others called it an acid-fast bacteria (which mutated into a fungus) and one called it an amoeba (e.g. trichomonad). Which of these is correct? Probably all of them ... Inside each cell are mitochondria. These mitochondria are where the energy of the cell is created, in the form of a molecule called ATP. The chemical process by which ATP are made start with what is called "The Krebs Cycle" or the "Citric Acid Cycle." This cycle of chemical reactions leads to the creation of ATP ... In a cancer cell, the Krebs Cycle is broken. Since the ETC is a spin-off of the Krebs Cycle, it is broken also. The result of breaking the Krebs Cycle is that the energy in the cell (i.e. the number of ATP molecules) drops dramatically. The human cell is a very sophisticated living thing. (!!!) When the Krebs Cycle is broken, the cell is generally able to fix the cycle, thus restoring the energy in the cell. But with a cancer cell, the cell is not able to restore its Krebs Cycle ... Now, researchers have identified a link between the body's system for regulating iron and breast cancer. The discovery could help doctors predict the course of breast cancer disease and even help them decide the most appropriate treatment. Iron is a vital nutrient, but too much iron can be harmful. Researchers at Wake Forest University in Winston-Salem, North Carolina, looked at a certain protein that regulates iron and studied the amount of it in breast cancer cells. "And what we found is that a protein called ferroportin that removes iron from cells, that eliminates iron from cells, was markedly reduced in breast cancers when compared to normal breast cells. And in fact was most reduced in the most aggressive breast cancers," said Frank Torti, who led the research team … The predictive power of ferroportin is still just a laboratory finding, but Torti says it suggests a possible novel approach to cancer therapy … Wake Forest University professor Frank Torti was interviewed by the journal Science Translational Medicine, which published his findings in their online edition.

Excess Iron May Trigger Growth. Three AICR-funded researchers are studying iron and zinc to determine their roles in cancer prevention and treatment … Association between elevated iron levels and an increased risk of developing cancer … For example, many breast and prostate cancer patients experience a “dormant” or slow growing tumor that suddenly begins to grow rapidly and spread. Because the promoter of this change is not well understood, AICR awarded a grant to Andre Kajdacsy-Balla, M.D., Ph.D., Professor of pathology at the Medical College of Wisconsin, to study the possibility that iron may increase cancer cell aggressiveness. His research team has already exposed human breast cancer cell cultures to excess iron and observed higher cell growth rates. Dr. Kajdacsy-Balla thinks the conversion of iron creates free radicals that damage surrounding tissues and allow cancer to spread. “Antioxidants, like vitamin C, may reduce the number of free radicals created,” he says. Cancer Cells Deprived of Iron. “We made the unexpected observation that curcumin binds and isolates iron in cells and tissues,” says Dr. Torti. The process of binding and isolating a metal ion such as iron is known as chelation. Her group made this discovery after studying the effects of several natural products. How chelation of iron by curcumin may prevent cancer is unknown. It may involve the special need of tumor cells for iron. Although iron is essential for the body to function, cancer cells are avid for iron. It is required for their growth and DNA synthesis. In conditions of excess iron storage, the risk of liver cancer is greatly increased. Dr. Torti notes, however, that chelation is probably only a part of how curcumin affects tumors. “Studies have shown lots of partial answers to the question of why curcumin inhibits tumor growth. Its property of chelating iron is just another piece of the puzzle of how this simple molecule works.”.

Their metabolism runs at full speed and requires large amounts of micronutrients, particularly iron. However, high levels of iron in the cell lead to the production of extremely harmful free radicals … Using a molecular-biological trick, Gulow and colleagues succeeded in blocking the production of one of the iron storage proteins in lymphoma cells. This leads to a rise in the level of free, non-bound iron in these cells. The iron boosts the production of free oxygen radicals which cause oxidative stress and, thus, cause damage to the cancer cells and induce their death. Healthy cells with their low iron level, however, survive the treatment unharmed. The DKFZ researchers have already found evidence that this iron effect also works in other lymphomas. They are now investigating whether selective release of iron may be a suitable approach for developing a novel cancer treatment.

Around the same time, research began to show that the regulation of iron is a central function of the immune system, and that this seems to have evolved because iron is a basic requirement for the survival and growth of cells of all types, including bacteria, parasites, and cancer. The pioneer researcher in the role of iron in immunity believed that an excess of dietary iron contributed to the development of leukemia and lymphatic cancers. Just like lead, mercury, cadmium, nickel and other heavy metals, stored iron produces destructive free radicals ... Excess iron is a crucial element in the transformation of stress into tissue damage by free radicals. For about 50 years, it has been known that blood transfusions damage immunity, and excess iron has been suspected to be one of the causes for this. People who regularly donate blood, on the other hand, have often been found to be healthier than non-donors, and healthier than they were before they began donating.

Material in this section is for general information only. First, popular culture inappropriately idealizes ingesting lots of iron as symbolized by Popeye and Superman, "the man of steel" prompting many to reach for the iron without thinking twice. However, highly conditioned athletes typically have low levels of body iron while maintaining peak physical performance. The solution to this misperception rests with the popular media ... Excessive iron is mistakenly believed to have no ill effects ... Anemia may have various causes (!!!) and should be treated with iron only after iron deficiency is diagnosed by blood testing. Detection of iron deficiency without obvious blood loss must be evaluated carefully because it may be due to serious internal disease (!!!) ... Third, we are flooded with iron because of supplementation of manufactured foods (especially cereal grains) that has been public health policy in the United States for over 60 years. Whether iron supplementation has been successful in reducing iron deficiency is not at issue here. The point is that the vast majority of individuals that ingest foods supplemented with iron are not - and never will be - iron deficient. Such supplementation represents treatment for a condition they do not have, that is taken involuntarily (without consent) and that contributes to disease risk with aging. Concern about iron deficiency is justified but concern about iron excess is at least equally justified. It is reasonable to suppose that individuals dying of natural causes since implementation of involuntary iron therapy without iron deficiency have had their life expectancy reduced to some extent because of this practice. The solution is to eliminate "shotgun" iron supplementation for the problem of iron deficiency. Manufactured foods should be prepared with or without added iron and the packaging labeled accordingly to informed consumer selection. Iron deficiency is a serious condition that must be diagnosed properly, its cause eliminated if possible, and corrected by treatment after which iron therapy should be stopped ... All foods contain iron because iron is essential for life. However, food iron content and availability are highly variable. In general, iron in plants is poorly absorbed while iron in meat is readily absorbed ... Manufacturers must indicate clearly on the front of the package in bold lettering whether or not iron has been added. Additional information on management of dietary iron intake can be obtained from www.irondisorders.org and other patient advocacy groups concerned with iron excess ...Blood removal is a safe method for rapid and precise iron reduction. Iron loss is natural for menstruating women but no such natural source of blood loss exists in men. Studies of routine blood donors have shown improvement in health status with donation of two or three units per year. Iron lost from donating this amount of blood is very close to the amount of iron absorbed in excess of requirements per year in the average individual. Too many young men enrolling in the Army weren't getting enough vitamins and minerals, especially iron, in their diets. To produce hardier soldiers (and citizens), the government mandated that flour and bread be fortified with iron. One result of this public-health intervention was a decline in anemia, which is caused by iron deficiency. But there may be some less-fortunate consequences.
Questions remain about how iron could cause cancer, but Leo Zacharski is convinced of the connection. "Iron loves to react with oxygen," he says. Its affinity for oxygen is what makes it essential for proper cellular functioning. Extra iron, however, can produce molecules called free radicals, known to damage DNA, proteins, and lipids—a possible pathway to malignancy. So is it time to ditch a policy from a different era? When functional iron deficiency is present in patients receiving rHuEPO, iron repletion is already recommended based on indirect evidence and biologic inferences as literally stated in the American Society of Clinical Oncology/American Society of Hematology guidelines. The study by Auerbach et al 1 addresses this issue in a controlled trial for the first time and confirms that iron supplement should be given intravenously rather then orally, as the latter is shown largely ineffective in cancer patients as it also occurs in the setting of anemia in other chronic diseases. A major point that remains to be elucidated in the field of rHuEPO therapy for anemia of cancer is whether iron supplement is capable of increasing the fraction of patients without iron deficiency responding to erythropoietic agents. Owing to the increased erythropoietic activity stimulated by rHuEPO, adequate delivery of iron to the bone marrow from the beginning of rHuEPO therapy may well optimize response in these patients as well (ie, by avoiding the development of iron deficiency, whose occurrence during rHuEPO therapy represents a major limitation to its efficacy). This important point should be addressed in future trials.

DKFZ (Deutsches Krebsforschungszentrum) is the largest biomedical research institute in Germany and is a member of the Helmholtz Association of National Research Centers. More than 2,200 staff members, including more than 1,000 scientists, are investigating the mechanisms of cancer (Krebs). Im April 1997 fand am Deutschen Krebsforschungszentrum in Heidelberg eine Konferenz mit internationaler Beteiligung über Spontanremissionen in der Onkologie statt. http://www.synergetik.net/spontanheilung.html Die Vorträge umfaßten ein weites thematisches Spektrum, wie etwa die Dokumentation und Epidemiologie von Spontanremissionen, grundlagenwissenschaftliche, psychologische und klinisch-onkologische Aspekte (Heim u. Schwarz, 1998a). Die Tagung war gut besucht und fand auch in den Medien und der Öffentlichkeit starke Beachtung. Dies überrascht nicht, denn abgesehen davon, daß eine genauere Kenntnis des Phänomens „Spontanremission“ nicht nur für wissenschaftlich tätige Onkologen, sondern auch für Krebspatienten von Interesse ist, haftet Spontanremissionen immer auch etwas Mysteriöses, Spektakuläres an. Eine methodisch fundierte Ursachenforschung wird durch die Seltenheit und Heterogenität der Fälle extreme erschwert und existiert bis heute nicht. Wir wollen im folgenden auf die Definition von Spontanremission zurückgreifen, die Everson u. Cole (1966) in ihrer Monographie gegeben haben und die nach wie vor weithin akzeptiert ist und benutzt wird. Hiernach ist eine Spontanremission “The partial or complete disappearance of a malignant tumour in the absence of all treatment, or in the presence of therapy which is considered inadequate to exert significant influence on neoplastic disease.”. Der Tumorrückgang und die Tatsache, daß dieser nicht therapiebedingt ist. Hingegen fordern Everson und Cole in ihrer Definition nicht, daß die Remission vollständig sein oder eine längere Zeit anhalten muß. Auch wird nicht unterstellt, daß überhaupt keine externen Ursachen für die Remission bekannt sein oder vermutet werden dürfen. Man kann hoffen oder erwarten, daß das Studium von Spontanremissionen das Verständnis für Krankheitsprozesse und biologische Mechanismen der Hemmung des Krankheitsprozesses fördert und zu vielversprechenden Forschungs- und Therapieansätzen führt. So gehen einige immuntherapeutische Behandlungsansätze letztlich auf frühe Beobachtungen von Spontanremissionen nach schweren Infekten zurück (Hirshberg, 1998). Zweitens kann die Existenz von Spontanremissionen für die Psyche des Patienten von Bedeutung sein. Die Existenz dieses Phänomens kann, selbst bei „austherapierten“ Fällen, zur Hoffnung Anlaß geben, daß es dem Körper allein gelingen könnte, Heilung oder Besserung herbeizuführen. Angesichts der Seltenheit von Spontanremissionen ist dieses Argument freilich problematisch, denn eine unrealistische Einschätzung der Heilungsaussichten kann die Auseinandersetzung mit der Krankheit und dem Tod auch erschweren. Dennoch trägt die Möglichkeit einer Spontanremission zur Unsicherheit der Prognose bei, und dieses Nichtwissen um das Lebensende kann, wie Gallmeier (1998) treffend bemerkt, durchaus Hilfe und Trost bedeuten. Einige Ereignisse, die vor Spontanremissionen beobachtet und als mögliche Ursache angesehen wurden: 1. Bakterielle oder virale Infektionen. 2. Operative Eingriffe oder Bestrahlung des Primärtumors. 3. Hormonelle Veränderungen (z.B. Schwangerschaft, Menopause). 4. Bluttransfusionen. 5. Elimination von Karzinogenen. 6. Veränderungen in der Psyche oder im Verhalten. Zusammenfassung. Spontanremissionen sind reale Phänomene, deren Erforschung für die Grundlagenforschung, die Beurteilung von Kasuistiken über Therapieerfolge und für die Auseinandersetzung von Patienten mit ihrer Erkrankung wichtig ist.

Spontaneous regression is rare biological event. It is not known, however, how often this remarkable phenomenon occurs and studies of spontaneous regression face specific challenges. Instances of spontaneous regression or remission (improvement or recovery without clear medical cause) are often documented in the cancer literature, with approximately 20 cases reported each year. Despite the identification of many individual cases, however, the frequency of this important phenomenon is poorly understood. While it is often quoted that spontaneous regression occurs no more than once in every 60,000 to 100,000 people with cancer. Spontaneous regression is said to occur when a malignancy partially or completely disappears without medical treatment or as a result of therapy that is considered inadequate to exert a significant influence on neoplastic disease.

American Cancer Society, Iron Supplementation and Cancer Anemia.
Unresponsiveness to erythropoiesis-stimulatory agents (ESA), which occurs in 30% to 50% of patients, is a major limitation to the treatment of chemotherapy-related anemia (CRA). This may be related in part to the dysregulation of iron metabolism, leading to functional iron deficiency. However, the use of iron supplementation during treatment with ESA has not been pursued as rigorously in anemic patients with cancer as it has in patients with chronic kidney disease. In this article, the authors discuss the role of iron supplementation in the setting of CRA in view of recent reports that have addressed this issue. In oncology, surprisingly, the use of iron supplementation during treatment with ESA has not been rigorously as applied in anemic patients with cancer as it has for dialysis patients. This underuse is caused at least in part by 1) the false perception that cancer patients do not have decreased iron stores (as measured by serum ferritin) and thus the belief that they do not to require iron supplementation during ESA therapy; 2) a failure to appreciate the relative blockade in access to storage iron pools or dietary iron that frequently exists in patients with chronic inflammatory illnesses such as cancer; and 3) the misinformation and misinterpretation of the incidence and clinical nature of serious adverse events of iv iron. In fact, parenteral iron still is considered a poorly tolerated medication that not infrequently harbors anaphylactic reactions. This opinion is erroneous and stems in part from the use of high molecular weight iron dextrans that, for a long time, were the only products available. Currently available iron preparations (low molecular weight iron dextran, iron saccarate, and ferric gluconate) are safe, as demonstrated clearly in dialysis patients, in whom the incidence of severe adverse events is extremely rare.
Patients with an absolute iron deficiency should receive iv iron supplementation only; until iron parameters are normalized, the administration of ESA would be ineffective.
Intravenous iron supplementation should be given when ESA therapy is instituted both in patients with FID and in iron-replete patients ... Patients with iron overload should not receive iron to avoid toxicity from iron excess.

“The carcinogenic potential of iron in colorectal cancer (CRC) is not fully understood. Population studies indicate an association between high dietary iron intake and CRC risk. Colorectal cancer (CRC) is the second most common cancer in developed countries. Apart from genetic mutations, environmental factors appear to play a role in intestinal carcinogenesis. Results from numerous population studies support the idea that dietary iron and/or elevated iron levels increase the risk of cancers including CRC, hepatocellular carcinoma (HCC) and lung cancer. HCC occurs at a higher incidence in hereditary haemochromatosis (HH) patients with hepatic iron overload than in the normal population and recently, an increased risk for CRC and breast cancer development in patients with HH has also been demonstrated. Further support for a role of iron in carcinogenesis comes from animal studies. Multiple injections of iron compounds, such as iron dextran complex, ferric nitriloacetate and ferric saccharate in rodents result in the formation of sarcomas, renal cell carcinoma and mesothelioma. Iron has also been implicated in intestinal carcinogenesis in rodent models of CRC. Hypothesis: high iron levels may potentiate the risk of cancer. Iron is a source of sustenance for cancer cell growth and proliferation. Cancer cell growth is enhanced by iron administration and has been shown to be retarded by both dietary iron deprivation and treatment with iron chelators.”.
Australian cancer researchers use iron compounds for formation of sarcomas, renal cell carcinoma and mesothelioma in rodents. NCI, ASCO and ACS recommended (in 2008-2009) iron compounds for treatment of American cancer patients. Cancer cells like iron and iron compounds (see iron-cancer researches of AICR and Norris Cotton Cancer Center). Cancer cells create cancer anemia. Modern anticancer poisons create supplementary anemia. NCI, ASCO and ACS suppress cancer anemia and supplementary anemia by means of iron compounds. American cancer patients are not rodents.

A person unable to have a balanced iron-rich diet may suffer from some degree of iron-deficiency anemia. 2. Body changes. An increased iron requirement and increased red blood cell production is required when the body is going through changes such as growth spurts in children and adolescents, or during pregnancy and lactation. 3. They identified HIV Rev Binding Protein (Hrb) as critically important for the development of T-ALL in two different mouse models. They discovered that Hrb accelerated the uptake of iron in the cancer cells, and this was a key mechanism behind its ability to enhance survival of the cells. Animal experiments show that mice that had reduced expression of Hrb had reduced T-ALL, and that iron-deficient mice developed Notch1-induced T-ALL significantly slower than normal. “A new therapeutic strategy is needed because current treatments that include high-intensity chemotherapy and cranial radiation are unsatisfactory, as they frequently cause severe long-term toxicities. Our results strongly implicate iron metabolism in the process of oncogenesis,” Richard Bram says.

The depression of serum iron levels in the presence of normal or slightly increased storage iron in cancer has its analogy in acute and chronic infections but the mechanisms responsible for the phenomenon is not understood. Chemical analyses of various malignant tumors described in the literature have demonstrated that the concentration of iron in such tissues is low. It is of importance to know whether or not ferromagnetic material might exist in human tissue … one might expect to find small amounts of magnetite in all animals, including humans. In an attempt to partially answer this question, I searched for magnetic remanence in four intact human adrenal glands which had been removed during autopsy and were frozen quickly in non-magnetic containers. Results of this analysis: indeed, there is a measurable amount of high-coercivity ferromagnetic material present which appears to be finely disseminated throughout the tissue. Between 1 and 10 million single-domain magnetite crystals per gram would be necessary to account for the observed magnetic remanence. Although these measurements do not uniquely identify the crystal phase as magnetite (ferromagnetic mineral) … Positive identification, of course, awaits the development of magnetic separation techniques capable of isolating and purifying these submicroscopic crystals. The presence of ferromagnetic material in human adrenal tissue suggests that it might be found in other organs as well. [The ferromagnetic material was present in the tissues before the magnetic analysis. Surgical contamination during autopsy was unlikely as the gland was removed intact and the tissue for analysis dissected from the interior with non-magnetic tools. Demagnetization is the process of reducing or removing the magnetism of a ferromagnetic material.
